In a recent episode, namely the "Behind the Laughter" episode, they actually mentioned the location of Springfield. Towards the end of the episode, when the voice over guy is talking, he mentions something about this "Northern Kentucky" town.
Also, there is actually a town called Shelbyville. I think it might even be in Kentucky somewhere.

There is a town named Shelbyville that is direcly adjacent to Springfield, Illinois. It is also worth mentioning that the shrine to Jebediah is similar to that for Lincoln in Springfield, Illinois.

Keep in mind, the crator, Matt Groenig, is from Oregon, and there is a Springfield Oregon. Many of his charecters and locations are named after Oregon locations
