My favorite "Guide" moment has to be the awakening to consciousness of the Whale. The internal monologue as the Whale comes to grips with its surroundings is a work of absolute genius. (The addition of the flowers' simple comment, which introduces an entirely new character connected to Arthur Dent, further enshrines this section of the "Guide" as the epitome of Mr. Adams' creativity.)
