Hiow do we know what we don't know?

Have just read the main page article on Conceptual tables. The bit about the periodic table was fascinating particulalry in that it indicated gaps in knowledge.
Remembering my statistical training for six sigma I also recall methods of calculus that 'put out feelers' in gray areas to provide indications where data should be.
There must be other methods out there too
Trouble is until every method has been evaluated how can we guage how big are the gaps in mankinds knowledge?
