One traditional way is to knock on the stone wall with the hilt of your sword. If it echoes, then there may be a hidden passage behind it.
You could also use infrared photography to track if any human has used the route recently. What use is a hidden stairway unless the person you are chasing has just gone through it?

Also, look for disturbed dust tracks, footsteps and worn floors, and smudges where they shouldn't be... (I watch far too much silly movies)

So many excellent suggestions! To think that for the last couple months I have been fruitlessly scouring city walls with a cup to my ear, when there were all these amazing super-spy techniques just waiting here to be discovered! Now I'm off to Google Earth to find out if any of these intriguing "yellow footprints" can be seen on Street View.

You could try turning off all the lights, and look to see if any light escapes from the other side of the secret room.
